Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001174089.2(SLC4A11):c.1849_1849+1del, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the SLC4A11 gene (transcript NM_001174089.2) at coding-DNA position 1849 through the canonical splice donor site of the intron immediately after coding-DNA position 1849, deleting this region. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. This variant is also known as c.1897_1897+1del. This variant has not been reported in the literature in individuals affected with SLC4A11-related conditions. This variant is present in population databases (no rsID available, gnomAD 0.007%). This sequence change creates a premature translational stop signal (p.Met633*) in the SLC4A11 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in SLC4A11 are known to be pathogenic (PMID: 17220209, 17679935).
