This variant lies in the PRDM16 gene (transcript NM_022114.4) at the canonical splice acceptor site of the intron immediately before coding-DNA position 3697, where A is replaced by G; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This sequence change falls in intron 16 of the PRDM16 gene. It does not directly change the encoded amino acid sequence of the PRDM16 protein. It affects a nucleotide within the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098).
