Pathogenic for Wilms tumor 1; Drash syndrome; 11p partial monosomy syndrome; Frasier syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_024426.6(WT1):c.1299T>A (p.Cys433Ter), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the WT1 gene (transcript NM_024426.6) at coding-DNA position 1299, where T is replaced by A; at the protein level this means converts the codon for cysteine at residue 433 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with WT1-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Cys428*) in the WT1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in WT1 are known to be pathogenic (PMID: 15150775).
