Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_002470.4(MYH3):c.1002C>T (p.Asp334=), citing LabCorp Variant Classification Summary - May 2015: Variant summary: MYH3 c.1002C>T (p.Asp334Asp) alters a conserved nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 7.6e-05 in 251284 control chromosomes. This frequency is not significantly higher than estimated for disease-causing variants in MYH3, allowing no conclusion about variant significance. c.1002C>T has been observed in one individual affected with autism as a de novo variant. These report(s) do not provide unequivocal conclusions about association of the variant with MYH3-Related Disorders. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
