Uncertain significance for Neuropathy, hereditary sensory and autonomic, type 2A; Pseudohypoaldosteronism type 2C — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_018979.4(WNK1):c.7110del (p.Ile2371fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the WNK1 gene (transcript NM_018979.4) at coding-DNA position 7110, deleting one base; at the protein level this means shifts the reading frame starting at isoleucine residue 2371, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change is expected to alter the c-terminus of the WNK1 protein (p.Ile2623Serfs*19). However, it is currently unclear if variants that occur in this region of the gene cause disease. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with WNK1-related conditions. ClinVar contains an entry for this variant (Variation ID: 2029933). Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
