Pathogenic for Congenital myasthenic syndrome — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_173660.5(DOK7):c.379G>A (p.Gly127Ser), citing LabCorp Variant Classification Summary - May 2015: Variant summary: DOK7 c.379G>A (p.Gly127Ser) results in a non-conservative amino acid change located in the IRS-type PTB domain (IPR002404) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-05 in 250258 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in DOK7 causing Congenital Myasthenic Syndrome (4e-05 vs 0.0014), allowing no conclusion about variant significance. c.379G>A has been reported in the literature in three homozygous individuals in a consanguineous family affected with Congenital Myasthenic Syndrome (Alsallum_2021). These data indicate that the variant is very likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
