Submitter rationale: Variant summary: TTN c.65120A>T (p.Lys21707Ile) results in a non-conservative amino acid change located in the A-band region of the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 4.6e-05 in 218654 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in TTN causing Cardiomyopathy, allowing no conclusion about variant significance. c.65120A>T has been reported in the literature in individuals affected with Dilated or Hypertrophic Cardiomyopathy (Mazzarotto_2020, Minoche_2019, Lopes_2013).
