This variant lies in the MYH2 gene (transcript NM_017534.6) at the canonical splice donor site of the intron immediately after coding-DNA position 4371, deleting one base. Submitter rationale: This variant has not been reported in the literature in individuals affected with MYH2-related conditions. This variant is also known as c.4371+1del. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. For these reasons, this variant has been classified as Pathogenic. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Ile1458Serfs*33) in the MYH2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in MYH2 are known to be pathogenic (PMID: 20418530, 23388406, 24193343).
