Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001174096.2(ZEB1):c.400C>T (p.Gln134Ter), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with ZEB1-related conditions. This sequence change creates a premature translational stop signal (p.Gln133*) in the ZEB1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in ZEB1 are known to be pathogenic (PMID: 16252232, 17935237, 30851240). This variant is present in population databases (rs772364498, gnomAD 0.003%).
