NM_000168.6(GLI3):c.3147C>A (p.Tyr1049Ter) was classified as Pathogenic for Pallister-Hall syndrome; Greig cephalopolysyndactyly syndrome by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the GLI3 protein in which other variant(s) (p.Gln1503*) have been determined to be pathogenic (PMID: 30235038). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. This variant has not been reported in the literature in individuals affected with GLI3-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Tyr1049*) in the GLI3 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 532 amino acid(s) of the GLI3 protein.
