Pathogenic for Capillary malformation-arteriovenous malformation syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002890.3(RASA1):c.2625del (p.Cys876fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the RASA1 gene (transcript NM_002890.3) at coding-DNA position 2625, deleting one base; at the protein level this means shifts the reading frame starting at cysteine residue 876, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This variant is not present in population databases (gnomAD no frequency). For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. This variant has not been reported in the literature in individuals affected with RASA1-related conditions. This sequence change creates a premature translational stop signal (p.Cys876Valfs*16) in the RASA1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in RASA1 are known to be pathogenic (PMID: 24038909).
