This variant lies in the TTN gene (transcript NM_001267550.2) at coding-DNA position 81337, where G is replaced by T; at the protein level this means converts the codon for glutamic acid at residue 27113 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The Glu25472Stop mutation in the TTN gene has not been reported previously as a disease-causing mutation or as a benign polymorphism, to our knowledge. Glu25472Stop is predicted to cause loss of normal protein function either due to production of an abnormal, prematurely truncated protein, or by absence of protein product due to nonsense-mediated mRNA decay. Other truncating TTN variants have been reported in approximately 3% of control alleles (Herman D et al., 2012). However, Glu25472Stop is located in the A-band region of titin, where the majority of truncating mutations associated with DCM have been reported (Herman D et al., 2012). Furthermore, Glu25472Stop was not observed in approximately 6000 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The variant is found in DCM panel(s).
