This variant lies in the TTN gene (transcript NM_001267550.2) at coding-DNA position 64999, where C is replaced by T; at the protein level this means converts the codon for arginine at residue 21667 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This sequence change creates a premature translational stop signal (p.Arg21667*) in the TTN gene. While this is not anticipated to result in nonsense mediated decay, it is expected to create a truncated TTN protein. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the gnomAD database. This premature translational stop signal has been observed in individuals with clinical features of dilated cardiomyopathy (PMID: 32880476, 36264615; internal data). ClinVar contains an entry for this variant (Variation ID: 202399). This variant is located in the A band of TTN (PMID: 25589632).
