Likely pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001220.5(CAMK2B):c.441G>C (p.Lys147Asn), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces lysine, which is basic and polar, with asparagine, which is neutral and polar, at codon 147 of the CAMK2B protein (p.Lys147Asn). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with epilepsy with intellectual disability and developmental delay (Invitae). In at least one individual the variant was observed to be de novo. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic.
