This variant lies in the CHEK2 gene (transcript NM_007194.4) at coding-DNA position 893, duplicating one base; at the protein level this means converts the codon for tyrosine at residue 298 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with CHEK2-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site.
