NM_000038.6(APC):c.170A>T (p.Asp57Val) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D57V variant (also known as c.170A>T), located in coding exon 2 of the APC gene, results from an A to T substitution at nucleotide position 170. The aspartic acid at codon 57 is replaced by valine, an amino acid with highly dissimilar properties. This alteration was detected in a cohort of 224 unrelated Brazilian individuals with breast cancer (Sandoval RL et al. PLoS One, 2021 Feb;16:e0247363). This amino acid position is well conserved in available vertebrate species. In addition, in silico predictors for this gene do not accurately predict pathogenicity. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
