Uncertain significance for Deafness-lymphedema-leukemia syndrome; Monocytopenia with susceptibility to infections — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_032638.5(GATA2):c.1267del (p.Glu423fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the GATA2 gene (transcript NM_032638.5) at coding-DNA position 1267, deleting one base; at the protein level this means shifts the reading frame starting at glutamic acid residue 423, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Glu423Argfs*54) in the GATA2 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 58 amino acid(s) of the GATA2 protein. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. This variant has not been reported in the literature in individuals affected with GATA2-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
