Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001134363.3(RBM20):c.1867C>T (p.Arg623Trp), citing Ambry Variant Classification Scheme 2023: The p.R623W variant (also known as c.1867C>T), located in coding exon 8 of the RBM20 gene, results from a C to T substitution at nucleotide position 1867. The arginine at codon 623 is replaced by tryptophan, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
