Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002528.7(NTHL1):c.664dup (p.Trp222fs), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the NTHL1 protein in which other variant(s) (p.Gln287*) have been determined to be pathogenic (PMID: 27329137, 28912133, 30753826; Invitae). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. ClinVar contains an entry for this variant (Variation ID: 2020093). This variant has not been reported in the literature in individuals affected with NTHL1-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Trp230Leufs*43) in the NTHL1 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 83 amino acid(s) of the NTHL1 protein.
