Likely pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_015021.3(ZNF292):c.1672C>T (p.Arg558Ter), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Arg558*) in the ZNF292 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 2166 amino acid(s) of the ZNF292 protein. This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with clinical features of ZNF292-related conditions (Invitae). In at least one individual the variant was observed to be de novo. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic.
