This variant lies in the AXIN2 gene (transcript NM_004655.4) at coding-DNA position 1786 through coding-DNA position 1834, deleting 49 bases; at the protein level this means shifts the reading frame starting at glutamic acid residue 596, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals affected with AXIN2-related conditions. Loss-of-function variants in AXIN2 are known to be pathogenic (PMID: 21416598, 15042511). However, tissue-specific alternative splicing of AXIN2 gene results in functional isoform lacking in-frame exon 7 (also known as exon 6, PMID: 15735151).
