Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_007055.4(POLR3A):c.2450C>A (p.Ser817Tyr), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the POLR3A gene (transcript NM_007055.4) at coding-DNA position 2450, where C is replaced by A; at the protein level this means replaces serine at residue 817 with tyrosine — a missense variant. Submitter rationale: This sequence change replaces serine, which is neutral and polar, with tyrosine, which is neutral and polar, at codon 817 of the POLR3A protein (p.Ser817Tyr). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with POLR3A-related conditions. ClinVar contains an entry for this variant (Variation ID: 2014854). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed at Invitae for this missense variant, however the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on POLR3A protein function. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
