Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_020812.4(DOCK6):c.2507T>A (p.Val836Asp), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals affected with DOCK6-related conditions. This sequence change replaces valine, which is neutral and non-polar, with aspartic acid, which is acidic and polar, at codon 836 of the DOCK6 protein (p.Val836Asp). This variant is present in population databases (rs755511381, gnomAD 0.006%). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
