This variant lies in the ANOS1 gene (transcript NM_000216.4) at coding-DNA position 1499 through coding-DNA position 1500, deleting 2 bases; at the protein level this means shifts the reading frame starting at valine residue 500, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change creates a premature translational stop signal (p.Val500Aspfs*27) in the ANOS1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in ANOS1 are known to be pathogenic (PMID: 8504298, 11297579). For these reasons, this variant has been classified as Pathogenic.
