This variant lies in the CAPN3 gene (transcript NM_000070.3) at coding-DNA position 2112 through the canonical splice donor site of the intron immediately after coding-DNA position 2115, deleting this region. Submitter rationale: In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic. This variant disrupts a region of the CAPN3 protein in which other variant(s) (p.Asp705Gly) have been observed in individuals with CAPN3-related conditions (PMID: 11297944, 25135358). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. This variant has not been reported in the literature in individuals affected with CAPN3-related conditions. This variant is not present in population databases (gnomAD no frequency). This variant results in the deletion of part of exon 19 (c.2112_2115+2del) of the CAPN3 gene. It is expected to disrupt RNA splicing.
