Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006204.4(PDE6C):c.1511C>T (p.Ala504Val), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The valine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals affected with PDE6C-related conditions. This sequence change replaces alanine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 504 of the PDE6C protein (p.Ala504Val). This variant is not present in population databases (gnomAD no frequency).
