NM_002137.4(HNRNPA2B1):c.265-75_614del was classified as Uncertain significance for Inclusion body myopathy with early-onset Paget disease with or without frontotemporal dementia 2 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the HNRNPA2B1 gene (transcript NM_002137.4) at 75 bases into the intron immediately before coding-DNA position 265 through coding-DNA position 614, deleting this region. Submitter rationale: This variant is not present in population databases (gnomAD no frequency). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals affected with HNRNPA2B1-related conditions. This variant results in the deletion of exons 5-6 and part of exon 7 (c.301-75_650del) of the HNRNPA2B1 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), however the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in HNRNPA2B1 cause disease.
