Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004656.4(BAP1):c.1359dup (p.Glu454fs), citing Ambry Variant Classification Scheme 2023: The c.1359dupA pathogenic mutation, located in coding exon 13 of the BAP1 gene, results from a duplication of A at nucleotide position 1359, causing a translational frameshift with a predicted alternate stop codon (p.E454Rfs*15). This variant has been observed in at least one individual with a personal and/or family history that is consistent with BAP1-related tumor predisposition syndrome (Ambry internal data). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
