Uncertain Significance for Long QT syndrome — the classification assigned by All of Us Research Program, National Institutes of Health to NM_000218.3(KCNQ1):c.1660G>A (p.Val554Met), citing ACMG Guidelines, 2015. This variant lies in the KCNQ1 gene (transcript NM_000218.3) at coding-DNA position 1660, where G is replaced by A; at the protein level this means replaces valine at residue 554 with methionine — a missense variant. Submitter rationale: This missense variant replaces valine with methionine at codon 554 of the KCNQ1 protein. Computational prediction suggests that this variant may have deleterious impact on protein structure and function (internally defined REVEL score threshold >= 0.7, PMID: 27666373). This variant is found within a highly conserved c-terminal cytoplasmic domain (a.a. 349-676). Rare non-truncating variants in this region have been shown to be significantly overrepresented in individuals with long QT syndrome (PMID: 32893267). To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with cardiovascular disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
