Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_144643.4(SCLT1):c.681del (p.Lys227fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Lys227Asnfs*7) in the SCLT1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in SCLT1 are known to be pathogenic (PMID: 28005958). For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with SCLT1-related conditions. This variant is not present in population databases (gnomAD no frequency).
