Likely pathogenic for T-B+ severe combined immunodeficiency due to JAK3 deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000215.4(JAK3):c.437T>G (p.Val146Gly), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces valine, which is neutral and non-polar, with glycine, which is neutral and non-polar, at codon 146 of the JAK3 protein (p.Val146Gly). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of severe combined immunodeficiency (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 2007667).
