Likely pathogenic for Neurodevelopmental disorder with or without hyperkinetic movements and seizures, autosomal dominant — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_007327.4(GRIN1):c.1931T>C (p.Val644Ala), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces valine, which is neutral and non-polar, with alanine, which is neutral and non-polar, at codon 644 of the GRIN1 protein (p.Val644Ala). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with GRIN1-related conditions (Invitae). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 2005963). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed at Invitae for this missense variant, however the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on GRIN1 protein function. In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic.
