Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_130837.3(OPA1):c.1133C>T (p.Thr378Ile), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt OPA1 protein function. This missense change has been observed in individual(s) with clinical features of OPA1-related conditions (Invitae). In at least one individual the variant was observed to be de novo. This sequence change replaces threonine, which is neutral and polar, with isoleucine, which is neutral and non-polar, at codon 323 of the OPA1 protein (p.Thr323Ile). This variant is not present in population databases (gnomAD no frequency).
