Pathogenic for Hereditary spastic paraplegia 63; Pontocerebellar hypoplasia type 9 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001368809.2(AMPD2):c.319C>T (p.Gln107Ter), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the AMPD2 gene (transcript NM_001368809.2) at coding-DNA position 319, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 107 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with AMPD2-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Gln161*) in the AMPD2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in AMPD2 are known to be pathogenic (PMID: 23911318).
