Likely pathogenic for Baraitser-winter syndrome 2; Autosomal dominant nonsyndromic hearing loss 20 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001614.5(ACTG1):c.188G>C (p.Gly63Ala), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine, which is neutral and non-polar, with alanine, which is neutral and non-polar, at codon 63 of the ACTG1 protein (p.Gly63Ala). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of ACTG1-related conditions (Invitae). In at least one individual the variant was observed to be de novo. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt ACTG1 protein function. In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic.
