Uncertain significance for Singleton-Merten syndrome 1; Aicardi-Goutieres syndrome 7 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_022168.4(IFIH1):c.2832del (p.Asn944fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the IFIH1 gene (transcript NM_022168.4) at coding-DNA position 2832, deleting one base; at the protein level this means shifts the reading frame starting at asparagine residue 944, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. This sequence change creates a premature translational stop signal (p.Asn944Lysfs*13) in the IFIH1 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 82 amino acid(s) of the IFIH1 protein. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with IFIH1-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
