This variant lies in the KCNH2 gene (transcript NM_000238.4) at 153 bases into the intron immediately after coding-DNA position 2398, where A is replaced by G. Submitter rationale: A variant of uncertain significance has been identified in an alternate transcript of the KCNH2 gene. The S851G variant has not been published as a pathogenic variant, nor has it been reported as a benign variant to our knowledge. This variant was not observed in approximately 3,400 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The S851G variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. However, this substitution occurs at a position that is not conserved across species, and Glycine is the wild-type amino acid at this location in at least two species. Additionally, 2/3 in silico algorithms predict this variant likely does not alter the protein structure/function.
