Uncertain significance for TNF receptor-associated periodic fever syndrome (TRAPS) — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001065.4(TNFRSF1A):c.1343C>G (p.Pro448Arg), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt TNFRSF1A protein function. This variant has not been reported in the literature in individuals affected with TNFRSF1A-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces proline, which is neutral and non-polar, with arginine, which is basic and polar, at codon 448 of the TNFRSF1A protein (p.Pro448Arg).
