Pathogenic for Marfan Syndrome/Loeys-Dietz Syndrome/Familial Thoracic Aortic Aneurysms and Dissections — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000138.5(FBN1):c.7205-1G>A, citing LabCorp Variant Classification Summary - May 2015. This variant lies in the FBN1 gene (transcript NM_000138.5) at the canonical splice acceptor site of the intron immediately before coding-DNA position 7205, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: Variant summary: FBN1 c.7205-1G>A is located in a canonical splice-site and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Several computational tools predict a significant impact on normal splicing: Four predict the variant abolishes the canonical 3' acceptor site. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 250774 control chromosomes. c.7205-1G>A has been reported in the literature in at-least two individuals affected with Marfan Syndrome and as a de-novo occurrence in at-least one case (example, Gezdirici_2020, Franken_2016). These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. One clinical diagnostic laboratory has submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation and classified the variant as likely pathogenic not citing the variant specific literature identified in the context of this evaluation. Based on the evidence outlined above, the variant was classified as pathogenic.
