This variant lies in the GDAP1 gene (transcript NM_018972.4) at coding-DNA position 1, deleting one base; at the protein level this means shifts the reading frame starting at methionine residue 1, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change affects the initiator methionine of the GDAP1 mRNA. The next in-frame methionine is located at codon 69. This variant is not present in population databases (gnomAD no frequency). For these reasons, this variant has been classified as Pathogenic. Disruption of the initiator codon has been observed in individuals with autosomal recessive Charcot-Marie-Tooth disease (PMID: 29184355, 33179230). It has also been observed to segregate with disease in related individuals.
