This variant lies in the FBN1 gene (transcript NM_000138.5) at coding-DNA position 4336, where G is replaced by A; at the protein level this means replaces aspartic acid at residue 1446 with asparagine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C15"). For these reasons, this variant has been classified as Pathogenic. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). ClinVar contains an entry for this variant (Variation ID: 200042). This missense change has been observed in individual(s) with clinical features of Marfan syndrome (PMID: 25907466; Invitae). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces aspartic acid, which is acidic and polar, with asparagine, which is neutral and polar, at codon 1446 of the FBN1 protein (p.Asp1446Asn). This variant also falls at the last nucleotide of exon 35, which is part of the consensus splice site for this exon.
