NM_000138.5(FBN1):c.3020T>G (p.Leu1007Arg) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L1007R variant (also known as c.3020T>G), located in coding exon 24 of the FBN1 gene, results from a T to G substitution at nucleotide position 3020. The leucine at codon 1007 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
