This variant lies in the GLB1 gene (transcript NM_000404.4) at coding-DNA position 69 through the canonical splice donor site of the intron immediately after coding-DNA position 75, deleting this region. Submitter rationale: This variant has not been reported in the literature in individuals affected with GLB1-related conditions. This variant is also known as c.69_75+1del. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. For these reasons, this variant has been classified as Pathogenic. This sequence change creates a premature translational stop signal (p.Gly23Glufs*8) in the GLB1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in GLB1 are known to be pathogenic (PMID: 18524657). This variant is not present in population databases (gnomAD no frequency).
