NM_004415.4(DSP):c.8330C>A (p.Pro2777His) was classified as Uncertain significance for Cardiomyopathy by Color Diagnostics, LLC DBA Color Health, citing ACMG Guidelines, 2015. This variant lies in the DSP gene (transcript NM_004415.4) at coding-DNA position 8330, where C is replaced by A; at the protein level this means replaces proline at residue 2777 with histidine — a missense variant. Submitter rationale: This missense variant replaces proline with histidine at codon 2777 of the DSP protein. Computational prediction suggests that this variant may have a deleterious impact on protein structure and function (internally defined REVEL score threshold >= 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in a child affected with long QT syndrome as well as in an unaffected parent (PMID: 27374306, 28784889). It has also been reported in an individual affected with left ventricular noncompaction (PMID: 28798025). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
