Submitter rationale: The p.L1995S variant (also known as c.5984T>C), located in coding exon 24 of the DSP gene, results from a T to C substitution at nucleotide position 5984. The leucine at codon 1995 is replaced by serine, an amino acid with dissimilar properties. This alteration has been reported in a sudden unexplained death cohort, as well as an arrhythmogenic right ventricular cardiomyopathy (ARVC) cohort (Hellenthal N et al. Europace, 2017 Nov;19:1881-1890; Marschall C et al. Cardiovasc Diagn Ther, 2019 Oct;9:S292-S298). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
