This variant lies in the POMT1 gene (transcript NM_001077365.2) at coding-DNA position 2141, where G is replaced by A; at the protein level this means converts the codon for tryptophan at residue 714 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This sequence change creates a premature translational stop signal (p.Trp736*) in the POMT1 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 12 amino acid(s) of the POMT1 protein. This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with muscular dystrophy-dystroglycanopathy (PMID: 22499106, 27193224). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 1998907). Algorithms developed to predict the effect of variants on protein structure and function are not available or were not evaluated for this variant.
