The arginine at codon 490 is replaced by cysteine, an amino acid with highly dissimilar properties. This variant has been detected in an arrhythmogenic right ventricular cardiomyopathy cohort; however, details were limited (Lin CY et al. J Cardiovasc Electrophysiol. 2019 Apr;30(4):582-592). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis.
