This variant lies in the DSG2 gene (transcript NM_001943.5) at coding-DNA position 562, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 188 with lysine — a missense variant. Submitter rationale: p.Glu188Lys (GAG>AAG): c.562 G>A in exon 6 of the DSG2 gene (NM_001943.3). The E188K variant has not been published as a mutation or as a benign polymorphism to our knowledge. The E188K variant was not observed in approximately 6,000 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The E188K variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is highly conserved across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. However, no missense mutations in nearby residues have been reported in association with cardiomyopathy, indicating this region of the protein may be tolerant of change.
