Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000023.11:g.108687484_108687501dup, citing Invitae Variant Classification Sherloc (09022015): This variant, c.4300_4317dup, results in the insertion of 6 amino acid(s) of the COL4A5 protein (p.Thr1434_Gly1439dup), but otherwise preserves the integrity of the reading frame. This variant is not present in population databases (gnomAD no frequency). This variant has been observed in individual(s) with clinical features of Alport syndrome (Invitae). In at least one individual the variant was observed to be de novo. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. For these reasons, this variant has been classified as Pathogenic.
