Pathogenic for Niemann-Pick disease, type C1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000271.5(NPC1):c.2170del (p.Leu724fs), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. For these reasons, this variant has been classified as Pathogenic. ClinVar contains an entry for this variant (Variation ID: 1997866). This sequence change creates a premature translational stop signal (p.Leu724Trpfs*5) in the NPC1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in NPC1 are known to be pathogenic (PMID: 9211850). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with NPC1-related conditions.
